Peptone water

Preparation of Culture Media (Peptone Water)

This medium is used for the testing of indole production, for the preparation of sugar media, and when highly alkaline (pH 8.0 - 8.4) is used for cultivation of Vibro cholerae.

• Peptone 10gm
• Sodium chloride 5gm
• Distilled water 1000ml

1. Dissolve by streaming (heat).
2. Adjust the pH to 7.5 (using 0.1M NaOH or 0.1M HCL).
3. Filter using filter paper.
4. Place into tubes or bottles.
5. Sterilise at 15lb & pressure for 20 minutes.
